Study of product effects on the production of collagen neo-synthesis in human fibroblasts.

Report: In Vitro Clinical Study

At the request of the INSTITUT D’EXPERTISE CLINIQUE – ESPAGNE, we have evaluated the effect of LIVING SILICA in the following parameters:

  • Synthesis of Collagen in Human Fibroblasts

  • Cytotoxicity in Human Fibroblasts

Evaluation of Cytotoxicity

The results are presented in the following table:

  Optical density %
  (570nm)
Negative Test 0.512 ± 0.03  -
Living Silica ® (0.2%) 0.508 ± 0.04 ns
Living Silica ® (0.5%) 0.500 ± 0.06 ns
Living Silica ® (1%) 0.514 ± 0.04 ns
 NS: Not Significant

The results show that LIVING SILICA does not present cytotoxicity to human fibroblasts in culture when used at 0.2%, 0.5%, and 1. %.

Evaluation of the Collagen Index

The separation and identification of hydroxyproline were performed by reversed-phase HPLC. Fluorescence peaks, after integration, used to calculate the concentration of hydroxyproline in the medical culture.

Concentration of hydroxyproline (mg/ml) % increase
Negative Test 1.64 ± 0.10  -
Living Silica ® 0.2% 1.79 ± 0.13 9
Living Silica ® 0.5% 1.86 ± 0.08 13
Living Silica ® 1% 1.95 ± 0.10 19

The results show that LIVING SILICA significantly increments the level of collagen produced by the human fibroblast cultures by comparison with the control.

Conclusion

In the experimental conditions adopted for the product LIVING SILICA :

  • Induces a significant increase in collagen levels of 13% and 19% respectively for the concentrations of 0.5% and 1% in cultured human fibroblasts compared with the control.

  • Does not induce cytotoxicity in the concentrations (0.2%, 0.5%, and 1%)

In conclusion, the LIVING SILICA product induces a significant increase in the index in cultured human fibroblasts.
